A note on sample size calculation for mean comparisons based on noncentral t-statistics.

چکیده
One-sample and two-sample t-tests are commonly used in analyzing data from clinical trials in comparing mean responses from two drug products. During the planning stage of a clinical study, a crucial step is the sample size calculation, i.e., the determination of the number of subjects (patients) needed to achieve a desired power (e.g., 80%) for detecting a clinically meaningful difference in the mean drug responses. Based on noncentral t-distributions, we derive some sample size calculation formulas for testing equality, testing therapeutic noninferiority/superiority, and testing therapeutic equivalence, under the popular one-sample design, two-sample parallel design, and two-sample crossover design. Useful tables are constructed and some examples are given for illustration.

Sample size and power calculations using the noncentral t-distribution
Abstract. The standard formulas for sample size and power calculation, as implemented in the command sampsi, make use of a normal approximation to the t-distribution. When the sample sizes are small, this approximation is poor, resulting in overestimating power (or underestimating sample size). One particular situation in which this is likely to be important is the field of cluster randomized t...
متن کاملOn the relative sample size required for multiple comparisons.
Multiple comparisons are commonly made in epidemiologic and genetic research. How to appropriately adjust for multiple comparisons remains a controversial issue. This note demonstrates, however, that large increases in the number of comparisons has a limited effect on the sample size required to maintain an experimentwise alpha-level. In particular, the relative sample size required increases o...
متن کاملComputing discrete mixtures of continuous distributions: noncentral chisquare, noncentral t
In this article, we address the problem of computing the distribution functions that can be expressed as discrete mixtures of continuous distributions. Examples include noncentral chisquare, noncentral beta, noncentral F , noncentral t, and the distribution of squared sample multiple correlation. We illustrate the need for improved algorithms by pointing out situations where existing algorithms...
متن کاملCalculation Methods for Wallenius' Noncentral Hypergeometric Distribution
Two different probability distributions are both known in the literature as "the" noncentral hypergeometric distribution. Wallenius' noncentral hypergeometric distribution can be described as an urn model with bias. Fisher's noncentral hypergeometric distribution is the conditional distribution of independent binomial variates given their sum. Several different methods for calculating probabili...

Sample size estimation is one of the crucial issues in the research methodology in medical sciences. It is an important issue for not only researchers but also readers of medical papers their frequent questions show how much they need to simple but accurate information. This paper presents the basic concepts of sample size calculation, and simplifies complicated issues using concrete example...
